The G2447A mutation does not affect ionization of a ribosomal group taking part in peptide bond formation.

Peptide bond formation on the ribosome is catalyzed by RNA. Kinetic studies using Escherichia coli ribosomes have shown that catalysis (>10(5)-fold overall acceleration) is due to a large part to substrate positioning. However, peptide bond formation is inhibited approximately 100-fold by protonation of a ribosomal group with pKa=7.5, indicating either a contribution of general acid-base cataly...

A transition-metal-free, one-pot procedure for the synthesis of α,β-epoxy ketones by oxidative coupling of alkenes and aldehydes via base catalysis.

A new strategy for the synthesis of epoxides is presented. This process allows the direct synthesis of epoxides from alkenes and aldehydes through C-H functionalization and C-C/C-O bond formation.

Synthesis of N-aryl-1-aminoindoles via intermolecular redox amination.

A redox amination strategy was developed for the synthesis of N-aryl-1-aminoindoles by N-N bond formation. Reaction of nitrosobenzenes with readily available indolines using Brønsted acid catalysis allows N-N bond formation under mild conditions. This method exploits the inherent reducing power of indoline to synthesize biologically relevant molecular architectures via redox amination. A one-po...

pKa cycling of the general acid/base in glycoside hydrolase families 33 and 34.

Glycoside hydrolase families 33 and 34 catalyse the hydrolysis of terminal sialic acid residues from sialyl oligosaccharides and glycoconjugates with a net retention of the stereochemistry at the anomeric centre. It is generally believed that the conserved aspartic acid in the active site functions as a general acid to protonate the hydroxyl group of the departing aglycone during glycosylation,...

Peptide bond formation on the ribosome: structure and mechanism.

The peptidyl transferase reaction on the ribosome is catalyzed by RNA. Pre-steady-state kinetic studies using Escherichia coli ribosomes suggest that catalysis (>10(5)-fold overall acceleration) is, to a large part, a result of substrate positioning, in agreement with crystal structures of large ribosomal subunits with bound substrate or product analogs. The rate of peptide bond formation is in...
